Abstract
The effects of quinine. chloroquine, primaquine and quinacrine on the uptake and incorporatioin of 14C-thymidine, 14C-uridine and 14C-amino acids were studied in synchronized Tetrahymena pyriformis. Quinacrine inhibited the incorporation of thymidine and uridimme but had little effect on the uptake of these precursors, which indicates that this antimalarial drug directly inhibits the synthesis of deoxyribonucleic acid (DNA) and ribonucleic acid (RNA) .The inihibitory effect of quinacrine on amino acid incorporation. however. appears to be due to an inhibition of uptake of these precursors. Quinine, chloroquine and primaquine inhibited the uptake of all precursors studied and this appears to be one of the mechanisms of the drugs for blocking precursor incorporation into DNA. RNA and protein.
