Abstract
1. A series of 5-phenyl, 5-R-hydantoins and related phenyl-R-acetylureas have been compared with respect to: (a) aqueous solubility; (b) minimal neurological toxic dose in rats; (c) ability to modify maximal electroshock seizure pattern in rats; (d) anti-Metrazol potency in rats; (e) ability to suppress induced hyperexcitability and hyperresponsiveness in frog sciatic nerve.
2. The data obtained indicate: (a) that all compounds showed some anti-convulsant potency except for the very insoluble diphenylacetylurea, and (b) that there is no close correlation between solubility, anti-electroshock potency, anti-Metrazol potency, and effects on peripheral nerve.
3. A comparison of potencies suggests that anticonvulsant activity of these substances does not require the formation of a common intermediate hydantoin or phenylacetylurea.
