A COMPARISON OF HEPATIC CYTOCHROME P450 PROTEIN EXPRESSION BETWEEN INFANCY AND POSTINFANCY
Section snippets
Patients and Methods
The human liver samples (except Nos 18 and 19) were collected postmortem from Japanese patients at the National Institute of Neuroscience after obtaining written informed consent from the patient's family. Two liver samples (Nos 18 and 19) were obtained from Japanese patients undergoing liver resection after obtaining written informed consent from the patient. Table 1 summarizes the sex, age, and causes of abortion, death, or surgery. Samples were obtained from portions without particular
Results
Representative Western blot results of sample Nos 4, 9, 17, and 19, and the reference standards are shown in Fig. 1. The contents of total CYPs determined spectrally and each CYP expressed in the liver of Perinatal/Infant Group and Control Group are summarized in Table 2. A good correlation was found between the contents of total CYPs measured spectrally and the sum of each CYP determined immunochemically (r=0.6764, p<0.005). The recoveries of sum of each CYP determined immunochemically
Discussion
We immunochemically measured the contents of each CYP protein expressed in the liver and compared them between the perinatal/infant patients (under 1 year old) and patients over 1 year old. Since previous studies on CYP1A2 showed lower expression and activity in the neonatal period 5, 6, 7, 8, we focused on CYP expression under 1 year of age and compared it to the control group in which samples were collected from patients at various ages so as to avoid a specific age bias. Although a good
