Structural biology of kainate receptors
2021, NeuropharmacologyCitation Excerpt :He found that dorsal root nerve fibers, which were strongly depolarized by kainate and the structurally related ligand domoate, showed low sensitivity to quisqualate, and were insensitive to NMDA (Agrawal and Evans, 1986; Watkins and Evans, 1981). However, the activation of native AMPA receptors in hippocampal neurons by both kainate and domoate, as well as by quisqualate, AMPA and glutamate, as demonstrated by kinetic analysis, revealed that kainate and domoate were not selective kainate receptor agonists (Patneau and Mayer, 1991). The subsequent cloning of glutamate receptor cDNAs revealed two subtypes of kainate receptor subunits, GluK1, GluK2 and GluK3, which form functional homomeric receptors in heterologous expression systems, and a second family, GluK4 and GluK5, originally named KA1 and KA2, that coassemble with GluK1-GluK3 to form heteromeric receptors with novel functional properties, but which do not form functional ion channels when expressed in isolation (Hollmann and Heinemann, 1994; Seeburg, 1993).
