Examples of the hydrolysis of d- and l-methylphenidate (d-MP and l-MP) by CES1A1, CES2, and CES3

d- and l-Methylphenidate (128 μM) were incubated with 100 μg/ml CES1A1, CES2, or CES3, respectively, in the total volume of 0.5 ml 20 mM Hepes buffer, pH 7.0, at 25°C for a 20 min or b 24 hours. This is a single experiment to demonstrate the enzymatic versus nonenzymatic hydrolysis of methylphenidate. Replicate kinetic analysis is shown in Table 3.
