Abstract
The vasodilatory effect of N, N'-bis[3'-(3'', 4'', 5''-trimethoxybenzoyloxy)-propyl] hexa-hydro-1,4-diazepine (a tertiary diamine) was studied on the cerebral and coronary vasculature of anesthetized dogs during measurement of blood flow and perfusion pres sure. Increases in cerebral (0.03-0.3 mg/kg) and coronary blood flow and vascular con ductance (0.01-0.3 mg/kg) occurred after i.v. injections, and these changes were ac companied with little or no effect on other cardiovascular and hemodynamic param eters. The coronary vasodilator responses in the dog and the chronotropic and inotropic responses in isolated guinea-pig atria to exogenous adenosine were markedly enhanced after exposure to the diamine. The high specificity of action for the cerebral and coronary vasculature, coupled with the known metabolic formation of adenosine solely in cardiac and brain tissue, suggests that the diamine possesses the capacity to act as a modulator of responsiveness of cerebral and coronary vascular smooth muscle to adenosine.
