Rapid long lasting PPARα activation in the spinal cord in the chronic Freund Adjuvant model of inflammatory pain [2] supports a role for PPARα signalling in the modulation of spinal nociceptive processing in models of chronic pain. It is well established that increased sensitization of spinal neurones, and activation of resident macrophage or glia cells, within the spinal cord are key mechanisms that contribute to aberrant pain responses in animal models of neuropathic pain [17,19].

All isoforms of PPAR were shown to reveal specific patterns of localization in different areas of the brain and spinal cord at neuronal and glial cell level [16,138,139]. Interestingly, PPAR-β/δ is widely distributed, while α and γ isotypes show a more constrained pattern of expression [16].

In a related model of spinal cord inflammation and chronic pain, PPARα binding to the DNA responsive elements was shown to occur (Benani et al., 2004). Oleic acid derivatives oleylethanolamide and 11-hydroxy-oleic acid are endogenous ligands of PPARα (Yokoi et al., 2010) and the application of oleic acid had neuroprotective effects in vitro, which depended on the activation of PPARα (Bento-Abreu et al., 2007).

The presence of PPAR-α in microglia suggests that this isoform modulates central inflammation, possibly by regulation of cytokine production by microglia (Benani et al., 2003, 2004). Interestingly, the levels of pro-inflammatory cytokine are significantly higher in the absence of functional PPAR-α (Genovese et al., 2005) which indicates the anti-inflammatory role of PPAR-α.
