Materials and methods
Animals. C57BL6/J mice were anesthetized with CO2 and decapitated. Olfactory mucosa was freshly dissected [20], frozen in liquid nitrogen, and stored at −80 °C.
Isolation of RNA. Total RNA was isolated using TRI REAGENT (Molecular Research, Cincinnati, OH) according to the manufacturer’s protocol. Total RNA concentrations were quantified spectrophotometrically at 260 nm.
RT-PCR. Tissue distribution of OAT6 was determined by RT-PCR. A panel of 6 mouse adult tissue cDNAs and 4 mouse fetal tissue
Identification of a novel SLC22 gene, OAT6, that is predominantly expressed in olfactory mucosa
The starting point for this study was the surprising observation of a novel putative organic anion transporter predominantly expressed in olfactory mucosa, a tissue about which little is currently known regarding the expression profile of OATs and OCTs. A novel murine sequence (ENST00000324623) located on chromosome 19 at position ∼6 Mbp was identified in the Ensembl mouse genome database by searching for novel genes annotated as slc22 family members (all data relate to the October 2003 version
