DHE has been shown to be oxidized by superoxide to form 2-hydroxyethidium or by non-specific oxidation by other sources of reactive oxygen species (ROS) to form ethidium [31]. Dihydrorhodamine 123 (DHR) is the most frequently used probe for measuring ONOO− [32]. This assay is based on the oxidative conversion of DHR123 to its corresponding two-electron oxidized fluorescent product, rhodamine.

    Citation Excerpt :

The oxidation of H2DCF to DCF can proceed in different manners. H2DCF oxidation can be mediated by various radical species (e.g., peroxyl, alkoxyl, NO2, carbonate [CO3−] and OH radicals and peroxynitrite hydroxyl and thiyl radicals that result from thiol oxidation) [54–58]. DCFH does not directly react with oxygen peroxide or O2–.
