Cellular distribution of γ-secretase subunit nicastrin in the developing and adult rat brains
2008, Neurobiology of AgingCitation Excerpt :To determine the possible co-localization of nicastrin and PS1 in the adult rat brain, we first established the normal distribution profile of PS1 immunoreactivity using a single labeling procedure and then performed double labeling experiments in sections from different brain regions. In agreement with earlier reports (Blanchard et al., 1997; Elder et al., 1996; Kim et al., 1997; Lah et al., 1997; Lee et al., 1996; Yan et al., 2004), PS1-immunoreactive neurons and fibers were distributed throughout the brain including the septal/diagonal band complex, nucleus basalis of Meynert, striatum, cerebral cortex, hippocampus, amygdala, thalamus, hypothalamus, median eminence, brainstem motor nuclei and cerebellar Purkinje cells (Figs. 5 and 6). Dual-labeling experiments revealed that nicastrin is co-localized with PS1-positive neurons in virtually all regions of the brain including vertical and horizontal limbs of the diagonal band complex (Fig. 5B and C), striatum (Fig. 5E and F), cerebral cortex (Fig. 5I and J), hippocampal formation (Fig. 5L and M), median eminence (Fig. 6B and C), thalamus (Fig. 6E and F), brainstem motor nuclei (Fig. 6H and I) and cerebellar Purkinje cells (Fig. 6K and L).

2007, Brain ResearchCitation Excerpt :Indeed, our immunolabeling study using an antibody targeting PS1 N-terminus (1–25 amino acid residues) shows intense labeling primarily in mossy fiber and olfactory nerve terminals. This laminar distribution of immunolabeling appears also more differential as compared to those displayed by antibodies targeting other domains of PS1 protein in either the olfactory bulb or hippocampus (Cribbs et al., 1996; Elder et al., 1996; Kim et al., 1997; Kodam et al., 2007). It is worthwhile noting that this and two of our recent studies using different radioligands and antibodies demonstrate that both Aβ-producing enzymes (BACE and γ-secretase) are particularly enriched in hippocampal mossy fibers and olfactory nerve terminals that form two of the most plastic synapses in the brain (Yan et al., 2004, 2007).
