Cohorts in previous studies of NMDARs in postmortem cortex consisted of 13–26 schizophrenia cases per group (Kristiansen et al., 2010; Simpson et al., 1991; Akbarian et al., 1996; Beneyto and Meador-Woodruff, 2008; Scarr et al., 2005; Woo et al., 2008a; Dean et al., 2016; Kristiansen et al., 2006; Kornhuber et al., 1989; Dracheva et al., 2001). Here, we investigated multiple NMDAR-related molecular indices in a cohort of 37 cases per diagnostic group (Catts et al., 2016), and we stratified by inflammatory status which increased the effect size in the smaller subgroups.

Postmortem study comparing normal controls to patients with schizophrenia revealed that schizophrenics have a reduced density of sigma binding sites in the frontal cerebral cortex (Simpson et al., 1991). There is evidence that activation of the PFC during the verbal fluency task, as assessed using multi-channel NIRS, is significantly lower in Pro carriers of the SIGMAR1 gene than in individuals with the Gln/Gln genotype (Takizawa et al., 2009a).

Postmortem human brain studies on brains from schizophrenic patients have indicated that the density of sigma receptors was lower in the temporal cortex (Weissman et al., 1991), but higher in the superior parietal cortex (Shibuya et al., 1992) than in the control tissue, although chronic neuroleptic treatment may contribute to these differences (Simpson et al., 1991; Hashimoto and London, 1994). Furthermore, it is also reported that [3H]nemonapride binding to sigma receptors in the caudate of postmortem brain of schizophrenic patients was significantly decreased as compared with control group (Helmeste et al., 1996).
