2017, Epilepsy ResearchCitation Excerpt :For electrophysiology studies, the microdisected hippocampal slices were equilibrated in ACSF at 24 °C on a mesh surface in a small beaker in a water bath (Thermo-Fisher Scientific, Waltham, MA) and continuously bubbled with oxygen (95% O2 and 5% CO2). In order to induce spontaneous epileptiform-like discharges, the hippocampal slices were perfused with ACSF-containing 4-aminopyridine (4-AP, 50 μM) (Avoli et al., 1996; Fueta and Avoli, 1992; Losi et al., 2016; Salazar et al., 2003). Once 4-AP was applied to the bath, the onset of spontaneous epileptiform spike activity occurred in ∼30 mins.
