Several reports have demonstrated that the serotonergic system play a role in many of the behavioral effects produced by nicotine (Damaj et al., 1994; Riekkinen et al., 1994; Suzuki et al., 1997; Seth et al., 2002). Nicotine (0.05 mg/kg) increased the levels of 5-HT and 5-HIAA but not NA, DA and DOPAC, in the DRN of wild-type mice.

Several reports have demonstrated that 5-hydroxytryptamine (5-HT) plays a role in many of the behavioural effects produced by NIC [42–44]. It is know that, after NIC administration, a decreased serotonergic firing in the DRN would elicit an anxiolytic-like response [45], whereas in the LS, increased 5-HT levels could result in anxiogenic-like effects [39].

It is reported that the anti-nociceptive effects of nicotine are reduced in a dose-dependent manner by serotonergic 5-HT1A agonists such as 8-hydroxy-2-(di-n-propylamino)tetraline (8-OH-DPAT) and buspirone [19]. Inhibition of serotonin biosynthesis and subsequent depletion of available serotonin stores enhances the anti-nociceptive effects of nicotine [20].
