The Gi/o-protein coupled receptor functions as a presynaptic autoreceptor that inhibits the release and synthesis of histamine or as a postsynaptic heteroreceptor that regulates other neurotransmitter systems (Arrang et al., 1995, 1983, 1987; Ligneau et al., 1998; Schlicker et al., 1989; Vanhanen et al., 2016). In particular, the ability of H3R to affect dopaminergic receptor expression and signaling via formation of complexes with dopamine D1 or D2 receptors in striatal medium spiny neurons has been well documented (Moreno et al., 2011, 2014; Rapanelli et al., 2016); also see reviews by (Bolam and Ellender, 2016; Ellenbroek, 2013).

The precise effects of histamine are exerted through stimulation of four different G-protein coupled receptor (GPCR) subtypes, namely H1-H4R (Fig. 4) [13–19]. All histamine receptors display, although with varying levels, constitutive activity [20], i.e. have an active conformation state that is independent of any ligand binding.
