2010, Sleep Medicine ReviewsCitation Excerpt :Interestingly, this A2A polymorphism has been associated with susceptibility to panic disorder134,135 and with the anxiogenic response to caffeine in healthy volunteers.136 In major depression, some evidences suggest a deficit in adenosinergic transmission,137,138 and studies indicate that adenosine could be implicated in mood regulation139–143 and in the antidepressant effects of sleep deprivation or of electroconvulsive therapy.144 REM sleep disinhibition observed in major depression but not in insomnia may directly relate to the monoamine hypothesis of depression that postulates a deficiency of noradrenaline (NA) and serotonin (5-HT) neurotransmission, because there are strong evidences that REM sleep generating neurons are under the opposite influence of inhibitory NA and 5-HT input and of excitatory cholinergic input.145,146

2008, Physiology and BehaviorCitation Excerpt :Moreover, the fact that caffeine counteracted the anti-immobility effect of amitriptyline in HI-rats but induced an antidepressant-like effect when applied alone suggests that endogenous adenosine can produce opposite behavioral effects depending on whether the 5-HT/noradrenergic transmission is enhanced or not. Previous studies have shown that TCA drugs inhibit adenosine uptake in brain cortical synaptosomes [52], explaining the observation that iontophoretically applied TCAs depress the firing rate of cortical neurons in vivo, and enhance the inhibitory actions of adenosine but not of an uptake-resistant analog [5]. The inhibitory effect of locally applied TCAs can be antagonized by systemic caffeine, suggesting that the increase of extracellular adenosine induced by these antidepressants is large enough to induce a significant activation of neuronal adenosine receptors [5].
