This confirms other studies that show a decline in motor function in aging rats (Altun et al., 2007; Bowenkamp et al., 1996; Ballak et al., 2014). We show for the first time in male Sprague–Dawley rats that motor neurons have a significant decline during aging.

Several studies have shown that GDNF enhances motor function in aged rats and non-human primates, which has been related to dopaminergic induction and regeneration of the nigrostriatal pathway (Bowenkamp et al., 1996; Emerich et al., 1996; Grondin et al., 2003; Herbert and Gerhardt, 1997; Lapchak et al., 1997b). In the present study, GDNF was over-expressed only in a restricted area of the hippocampus and the swimming speed was not affected.

2008, NeuroscienceCitation Excerpt :The motor function impairments observed on the beam walking task are comparable with published findings in which C57 BL6/J mice treated with acute and sub-chronic dosing regimens of MPTP, were reported to display impairments in limb coordination, stride length and motor function at 1–2 weeks post-MPTP administration (Ogawa et al., 1985; Fernagut et al., 2002). It seems that the nigrostriatal dopaminergic circuit has been implicated specifically in the successful completion of balance beam tasks (Bowenkamp et al., 1996). Qualitative analysis showed that Mn-exposed animals exhibit hind-limb weakness, delayed motor initiative (akinesia), postural instability, freezing behavior and action tremor; regarding these alterations, Autissier et al. (1982) reported that mice subchronically exposed to Mn by intragastric gavage showed hypoactivity, this change was associated with a drop in striatal DA of 50%; Erikson et al. (1987) found that about 5 months after the start of the Mn exposure the animals became hypoactive with an unsteady gait, and subsequently action tremor.

Other studies in normal aged rats have also demonstrated improved motor abilities [4,15,26], and increases in tyrosine hydroxylase (TH) activity in the substantia nigra and striatum [26], following GDNF administration. These studies indicate that the nigrostriatal DA system of normal aged rats is able to respond to GDNF.
