GABA withdrawal syndrome: GABA<inf>A</inf> receptor, synapse, neurobiological implications and analogies with other abstinences
2016, NeuroscienceCitation Excerpt :One-week administration of Flurazepam and abrupt interruption induces anxiety associated with increased expression of AMPA-type glutamate receptors and an increase in the activity of Ca2+-channel-dependent voltage in hippocampal neurons, reflecting neuronal hyperexcitability (Shen and Tietz, 2011; Earl et al., 2012; Cloos and Bocquet, 2013). Between 1971 and 1978, several cases were reported in the U.S. that were related with prolonged treatment with Diazepam and its abstinence: anxiety; insomnia; profuse sweating; dysphoria, agitation; emotional instability; tremor, headache; dizziness; lack of coordination; sensory hypersensitivity; fatigue; lethargy; blurred vision; facial burning sensation; muscle pain; tachycardia; hallucinations; psychosis; delirium and, sometimes, convulsions (Greenblatt and Shader, 1981; Doble and Martin, 1996; Tan et al., 2011; Arnot et al., 2001; Kovačević et al., 2014). In rats, interruption of prolonged treatment with Diazepam creates the appearance of irritability, fearful behavior, and seizures.
Modulation of native GABA<inf>A</inf> receptor activity by triazolo 1,5-benzodiazepines
2013, NeuroscienceCitation Excerpt :After their discovery 50 years ago (Sternbach et al., 1962) 1,4-benzodiazepines were soon demonstrated to act as anxiolytics (Zbinden and Randall, 1967; Doble and Martin, 1992).
Synthesis, pharmacological studies and molecular modeling of some tetracyclic 1,3-diazepinium chlorides
2010, Bioorganic and Medicinal ChemistrySynthesis of novel bioactive derivatives of 3-(4-chlorophenyl)-2-hydrazino-5,6,7,8-tetrahydrobenzo(b)thieno[2,3-d]py rimidine-4(3H)-ones
2009, European Journal of Medicinal ChemistryCitation Excerpt :The GABAB receptor belongs to the G-protein-coupled receptors super family, while the GABAA [2] and GABAC [3] are ligand gated chloride ion channel complexes. GABAA receptors, which are responsible for the majority of neuronal inhibition in the mammalian CNS, mediate the action of many pharmacological useful agents, including benzodiazepines, barbiturates, neuroactive steroids, anesthetics and convulsants [4,5]. At least two classes of compounds have been identified by their ability to modulate GABA neurotransmission by interacting with receptor complex.
