Abstract

A rapid microassay is described for the measurement of biogenic amines using an isocratic HPLC system with electrochemical detection. Catecholomines, indoleamines and their major metabolites were extracted with 150 μl of perchloric acid from brain tissue punches (<250 μg) using a simple one-step sample preparation method. These compounds were separated on a short (80 mm) column with 3 μm particle size packing, and electrochemically detected within a total run time of less than 6 minutes. Detection limit sensitivity was approximately 2–5 pg. This method, detailed in an easy-to-follow description, reduces assay time, minimizes the possibility for errors, maximizes efficiency, and requires only standard HPLC equipment and supplies.

Before sample analysis a solution was prepared with 1 ml of stock solution with 2 ml of sodium tetraborate 0.1 M. The pre-column derivatization was completed by reacting 100 μl of this solution with 50 μl amino acid standard solution for exactly 2 min before the injection in the analytic column (Donzanti and Yamamoto, 1988a,b). For this study, an isocratic HPLC system, a fluorescence detector, a sample injector for HPLC, an RP-18 50 × 4.6 mm and a degasifier were utilized.

Only rats with probes in the dorsal hippocampus were included in the data analysis. Concentrations of glutamate in microdialysis samples as well as vesicular glutamate content were analyzed via HPLC coupled to fluorescence detection as previously described (Donzanti and Yamamoto, 1988). Glutamate was derivatized with O-phthaldialdehyde (OPA; Sigma-Aldrich) prepared by dissolving 27 mg OPA in 9 mL of 0.1 M sodium tetraborate pH 9.4 and 1 mL of 100% methanol to which 15 μL of β-mercaptoethanol was added.
