For this study, an isocratic HPLC system, a fluorescence detector, a sample injector for HPLC, an RP-18 50 × 4.6 mm and a degasifier were utilized.

Concentrations of glutamate in microdialysis samples as well as vesicular glutamate content were analyzed via HPLC coupled to fluorescence detection as previously described (Donzanti and Yamamoto, 1988). Glutamate was derivatized with O-phthaldialdehyde (OPA; Sigma-Aldrich) prepared by dissolving 27 mg OPA in 9 mL of 0.1 M sodium tetraborate pH 9.4 and 1 mL of 100% methanol to which 15 μL of β-mercaptoethanol was added.
