It has been observed that the neuropathologic cellular changes during sustained epileptic activity in the hippocampus are characterized by an intense transient influx of calcium leading to mitochondrial functional impairments (Heinemann et al., 2002; Patel, 2002) and also by the triggering of intracellular enzyme cascades, including the activation of nitric oxide synthase, which leads to increased levels of reactive oxygen species (ROS) and oxidative stress (Farooqui et al., 2001; Lafon-Cazal et al., 1993). Formation of free radicals results in extensive lipid peroxidation, which damages cellular organelles and membranes, and finally leads to cell death.

2009, Brain ResearchCitation Excerpt :It is likely that protection by Cu,Zn-SOD was produced through an extracellular site, since it was protective when applied as co-treatment for only 1 h and at low activity (3 U/ml), whereas Cu,Zn-SOD is generally unable to penetrate cell membranes readily even at high concentration. This assertion arises from previous work in which treatment or pre-treatment for up to 24 h with Cu,Zn-SOD did not prevent glutamate-receptor mediated excitotoxicity in cultured neurons, even though an intracellular production of superoxide was clearly implicated in the toxicity (Patel et al., 1996; Lafon-Cazal et al., 1993). Similarly Cu,Zn-SOD, when pre-applied for 1 h at concentrations much greater than 3 U/ml (up to 100 U/ml), was unable to scavenge intracellular superoxide in bovine aortic endothelial cells (Zhao et al., 2005).
