The anti-inflammatory effect of kappa opioid agonists has been reported in inflammatory pain models in rats (Walker et al., 1995; Binder et al., 2001; Walker, 2003; Bileviciute-Ljungar et al., 2006) and in mice (Paton et al., 2017). Also, Bastos et al. (2011) showed that U-50,488 (a kappa opioid receptor agonist) reduced bone loss and IL-6 levels and increased IL-10 in ligature-induced periodontal disease in rats.

It is well demonstrated that, in addition to analgesia, κ-opioid receptor agonists, including peripherally selective agonists, display potent anti-inflammatory actions (Walker et al., 1995; Wie et al., 1998; Catheline et al., 1999; Binder et al., 2001). Therefore, a possible anti-inflammatory activity of crotalphine, which may contribute to its antinociceptive action, cannot be ruled out.
