A skin-sensitizing and thermolabile antibody in the mouse

Abstract

A skin-fixing, heat labile and “mast cell sensitizing” antibody was recently found in the rat by one of us (1, 2, 3, 4). The fact that the mouse stands phylogenetically close to the rat added to the recent observation of Prouvost-Danon $et al$ . (5) showing that the ability to release histamine from mouse peritoneal mast cell arises early after sensitization and disappears after a short time lead us to investigate the presence of a similar antibody in the mouse. The present paper reports the results of experiments showing that the mouse produces a skin-sensitizing and thermolabile antibody that is here provisionally called “mouse early antibody”.

The protective effects of two antihistamines and two anti-allergic drugs against anaphylactic paw edema were studied in immunized animals that had or had not received a booster injection of antigen. The injection of 1 or 10 μg/paw ovalbumin induced acute paw edema of similar intensity in both groups. The antihistamine meclizine and the mixed anti histamine/anti-5-HT antagonist cyproheptadine reduced the anaphylactic reaction by 55 and 84% respectively, in non-boosted animals and were less effective against edema induced by 1 μg antigen in boosted animals. The effectiveness of these drugs was also reduced when boosted mice were challenged with 10 μg antigen, where meclizine and cyproheptadine inhibited edema by 31 and 59%, respectively. The anti-allergic compounds ketotifen and azelastine, although effective against allergic inflammation in non-boosted mice, had a reduced or no effect in boosted mice. Our results suggest that allergic edema is less sensitive to antihistamine and anti-allergic drugs in boosted mice, which may be accounted for by an increased role of other mediators.
Action of pertussigen (pertussis toxin) on serum IgE and on Fcϵ receptors on lymphocytes

Pertussigen (pertussis toxin (PT)) is one of the most effective stimulators of IgE production in mice and rats. Employing flow microfluorimetric analysis (FMF), we showed that PT increases the percentage of blood and spleen lymphocytes with IgE on their surface. The percentage of IgE-bearing cells in the spleen of normal untreated C57B1/10SCN mice of various ages varied from 2.2 to 12.2%, with an average value of 6.1 ± 5.4%. In mice treated with 400 ng of PT and 1 mg of chicken egg albumin (EA), the percentage of these cells increased, 14 days after immunization, to an average value of 31.1 ± 2.2%. Immunization of mice with PT alone increase the percentage of IgE-bearing cells only slightly (13.1 ± 2.2% of the splenic lymphocytes) while injection of 1 mg of EA alone did not have any detectable action. As little as 6 ng of PT, when given simultaneously with 1 mg of EA, increased the percentage of IgE-bearing lymphocytes. A booster dose of 10 μg of EA given on Day 14 induced a further increase in the percentage of these cells even when as little as 0.039 ng of PT had been given at the time of initial immunization. PT was effective when given 4 days before or 5 days after EA. EA was effective when given 4 days before or 4 days after PT, but not 8 days after. The increase in IgE-bearing cells was mainly due to cytophilic binding of IgE to receptors for the ϵ chain of IgE (Fcϵ) on the surface of lymphocytes rather than to a greater number of IgE-producing cells. This was shown by removing the IgE from Fcϵ receptors by acid treatment which reduced the percentage of IgE-bearing cells to nearly normal values. The antibodies of IgE class with specificity to EA were increased dramatically, while antibodies with specificity to PT were not detected.

In three mouse strains, BALB/c, CBA and C3H/He, specific IgE antibodies could be induced by DNP-GAT and GAT. The presence of both anti-GAT and anti-DNP IgE antibodies was measured, using both heterologous PCA in rats and specific degranulation of peritoneal mast cell tests. The fine specificity of the IgE anti-GAT antibodies elicited by both DNP-GAT and GAT was similar to the specificity of anti-GAT IgG antibodies. The IgE anti-GAT antibodies cross reacted with l-glutamic acid⁵⁰-l-tyrosine⁵⁰ (GT) copolymer, but not with l-glutamic acid⁶⁰-l-alanine (GA) copolymer. Anti-idiotypic serum was found to be able to degranulate specifically peritoneal mast cells from GAT and DNP-GAT immunized animals, indicating that IgG and IgE anti-GAT antibodies share common idiotypic determinants.
