However, the use of NK1 antagonists had been correlated with the appearance of diarrhoea (Bubalo et al., 2018; Yuan et al., 2016), and no literature had evaluated the (Huber et al., 1993) stomach and colon at the same time in an emetic animal model to explain this side effects. It is important to note that NK receptors are expressed in smooth muscle, ICCs and neurons in the GI (Drokhlyansky et al., 2020; Lee et al., 2015; Lee et al., 2017; Liu et al., 2015).

Thus the contractile response to natural tachykinins or selective receptor agonizts resulted from activation of NK2, but not NK1, receptors (Huber et al., 1993b), and NK2 receptor blockade inhibited the largest proportion of tachykinin-evoked responses (Kovac et al., 2006). The transcript level of the TACR2β isoform was much less than that of the α isoform, but it was consistently more than that of TACR1 or TACR3.

The results are consistent with the previous reports demonstrating that NK-2R agonist induced vigorous contractions of muscle strip of the rat and mouse stomach and human lower esophageal sphincter and ileum [33–36]. Moreover, whereas L-732,138 had no effect on NKA-induced contraction, the NK-2R antagonist GR159897 significantly inhibited NKA-induced contractions in longitudinal muscles and GR159897 at 100 μM almost abolished contractile effect of NKA at 3 μM.
