The decay of deltamethrin-induced tail currents is voltage-dependent (Ruigt et al., 1987); at a more depolarized Vhold, the deactivation is slower, and thus the decay of tail currents is slower. Therefore, we hypothesized that increasing the Vhold would slow down the deltamethrin-induced tail currents in Nav1.8 and thereby cause a use-dependent increase in the percent of deltamethrin-modified Nav1.8 channels.

Interestingly, pyrethroids, a group of monoterpene esters used as insecticides, act primarily by activating, instead of inhibiting, neuronal (Ruigt et al., 1987; Du et al., 2013) and cardiac (Spencer et al., 2001; Spencer & Sham, 2005) Na+ channels (for reviews, Clark & Symington, 2007; Narahashi et al., 2007; Soderlund, 2012). The abovementioned effects of monoterpenes (except pyrethroids) on VGSCs have also shown to be associated with their analgesic and antinociceptive activities.
