Thiobarbituric acid value on fresh homogenate of rat as a parameter of lipid peroxidation in aging, CCl4 intoxication, and vitamin E deficiency

Abstract

The changes of thiobarbituric acid (TBA) values due to aging, CCl4 intoxication, and vitamin E deficiency were determined on fresh (nonincubated) tissue homogenate by the 1% H3PO4 method (19).

The TBA values of liver and testis increased with age in good correlation. The correlation coefficients of liver up to 42 weeks old, testis up to 16 weeks old, and testis up to 42 weeks old were 0.694, 0.906, and 0.775, respectively.

Administration of CCl4 caused a prolonged increase of liver TBA values as determined by the 1% H3PO4 method, although many reports have failed to detect them.

The significant elevation of TBA values of fresh tissues (liver, kidney, heart, lung, and testis) from rats fed a vitamin E-deficient diet for 18–19 weeks was obtained by the present method, which so far has been recognized only after an appropriate incubation of tissue preparation.

The TBA-reactive substances measurable by the present method include several substances other than water-soluble malonaldehyde, and may correlate well with the overall rancidification due to lipid peroxidation in biological tissues.
