2013, European Journal of PharmacologyCitation Excerpt :While FK506 and rapamycin increased, inhibition of calcineurin by cypermethrin, okadaic acid and calcineurin inhibitory peptide, failed to alter the potentiation of ryanodine receptor-mediated Ca2+ release by the immunosuppressive drugs, in either the colon or portal vein. Neither did cypermethrin nor okadaic acid alter [Ca2+]c in portal vein or coronary artery (Ashizawa et al., 1989; Hirano et al., 1989; MacMillan and McCarron, 2009), nor did cyclosporin A alter [Ca2+]c in pulmonary (Zheng et al., 2004) or coronary artery (Frapier et al., 2001), nor store [Ca2+] in aorta (Avdonin et al., 1999). Yet, the binding of calcineurin to the ryanodine receptor by itself appears to regulate the activity of the receptor in colonic smooth muscle, as revealed by increased ryanodine receptor-mediated Ca2+ release by the calcineurin inhibitor, cypermethrin (MacMillan et al., 2008).
