Selectins: critical mediators of leukocyte recruitment

Abstract

Selectins are multi-functional adhesion molecules that mediate the initial interactions between circulating leukocytes and the endothelium. First identified over a decade ago, selectins have provided insight into areas as diverse as normal lymphocyte homing, leukocyte recruitment during inflammatory responses, carbohydrate ligand biosynthesis and adhesion-mediated signalling. This review will examine the selectins and their ligands with a focus on recent findings using knockout technology as well as the emerging role of selectins as signalling molecules.

      During physiological or pathological inflammation and hemostasis, selectins initiate multicellular adhesive and signaling events, which emphasize the interactions between leukocytes and endothelial cells. As a result, the initiation of the leucocyte adhesion cascade is a multi-step process that occurs during constitutive homing of naive lymph nodes (Patel, Cuvelier, & Wiehler, 2002). The platelet-PSGL-1 interaction on myeloid cells starts the signaling by the selectin molecule.

      C-type lectins have been classified into 17 groups of vertebrates based on their structures and functions. Collectins are one such group, and mainly serve as PRRs to discriminate self and nonself [5,6], while another group, selectins, mediate cellular adhesion to activate encapsulation and inflammation [7]. Unlike the abyssal zone, which is characterized by low biomass and high biodiversity [8,9], submarine hydrothermal systems are endemism hotspots harboring high biomass and low biodiversity [10,11].
