Local Control of Granule Cell Generation by Cerebellar Purkinje Cells

Abstract

Cerebellar Purkinji cells were ablated by the specific expression of diphtheria toxin in these cells in transgenic mice. Purkinje cell degeneration during early postnatal development shows a zonally restricted pattern which has been exploited in order to look for local secondary effects. The most obvious early effect is the alignment of gaps in the Purkinje cell layer with dramatically thinned zones in the overlying EGL, the germinal layer from which granule cells are generated. Within these EGL zones in the transgenic mutant, markers that distinguish matrix from mantie cells demonstrate a preferential loss of the proliferative cells. Comparison of BrdU incorporation in the mutant vs wild-type confirms the reduction in proliferation. In the mutant, in situ labeling of DNA fragmentation associated with apoptotic cell death shows abundant labeling of granule cells that have exited the EGL, but not of progenitor cells In the EGL. Thus, although a trophic role for Purkinje cells has been well documented, these observations further suggest a mitogenic role which can be exerted locally.

    The Shh signaling pathway is crucial to the regulation of GCPs proliferation (Jiang et al., 2011; Varjosalo and Taipale, 2008), and sustains normal cerebellum foliation in the developing cerebellum (Lewis et al., 2004). Shh is a major mitogenic factor secreted by Purkinje cell, which is thought to primarily and actively regulate the amplification of GCPs proliferation in the EGL (Luca et al., 2016; Smeyne et al., 1995). The addition of Shh to GCPs results in a prolonged proliferation reaction, whereas GCP proliferation is inhibited by the down-regulation of Shh (Dahmane and Ruizialtaba, 1999; Lewis et al., 2004).

    In accordance with the native L7 expression, the transgenic line showed β-galactosidase expression solely in Purkinje cells, indicating that the 8-kb genomic fragment involves regions critical for Purkinje cell-restricted expression. Subsequently, Smeyne et al.23 generated transgenic mice expressing an attenuated form of the diphtheria toxin under the control of the 3-kb L7 fragment in which the cDNA was inserted into a unique BamHI site present in exon 4, whereas the original start codon in exon 2 and other potential ATGs in exons were mutated (see L7-1 in Figure 2A). The transgenic mice expressing a transgene using the L7-1 promoter exhibited highly specific GFP expression in Purkinje cells,16,23–25 which strongly indicates the presence of regions regulating the Purkinje cell-specific expression in the L7-1 promoter.
