It has been reported that intestinal PEPT1 is regulated by various factors [23], such as thyroid hormone [24], dietary conditions [25,26], diurnal rhythm [27] and a selective σ-ligand, pentazocine [28]. In addition, the ectopic induction of PEPT1 expression in the colon, where PEPT1 was not expressed under normal conditions, was observed in cases of functional deficiency of the small intestine such as ulcerative colitis, Crohn's disease and short-bowel syndrome [23].

The composition of the transport buffer was 25 mM HEPES/Tris (pH 7.4), 140 mM NaCl, 5.4 mM KCl, 1.8 mM CaCl2, 0.8 mM MgSO4, and 5 mM glucose (Fujita et al., 1999, 2005; Inoue et al., 2003; Tanaka et al., 2005; Wada et al., 2005). Following incubation, the buffer containing radiolabeled compound was aspirated off and the cells were washed twice 2 ml of ice-cold transport buffer.
