Monitoring DNA polymerase β mitochondrial localization and dynamics
2022, DNA RepairCitation Excerpt :Here, we provide evidence to support our hypothesis that nuclear-encoded pol β is associated with, and can be recruited to, mitochondrial nucleoids for repair of mtDNA. There has been a long-standing debate regarding a role for pol β in repair of mammalian mtDNA damage [22,23]. Two recent publications [24,25] provided clear evidence that pol β is present in mitochondria and mediates BER activity.
DNA polymerase β: A missing link of the base excision repair machinery in mammalian mitochondria
2017, DNA RepairCitation Excerpt :The Crithidia fasciculata mitochondrial pol β is capable of both of the BER enzymatic activities found in mammalian nuclear pol β, i.e., gap-filling and lyase. Third, Low and associates [37] identified pol β in bovine heart mitochondria in a series of biochemical experiments that are consistent with the results reported here. Also, a cross-linked protein consistent with pol β was observed in Percoll gradient purified mitochondria, but was considered a contaminant [65].
Impaired adaptive resynthesis and prolonged depletion of hepatic mitochondrial DNA after repeated alcohol binges in mice
2002, GastroenterologyCitation Excerpt :*Significantly different from control values (P < 0.05). N-ethylmaleimide, an inhibitor of most cell DNA polymerases including DNA polymerase γ,27 also had inhibitory effects, whereas aphidicolin, an inhibitor of nDNA polymerases α, δ, and ϵ but not mtDNA polymerase γ,27 did not (Figure 3A). We then compared mtDNA synthesis in hepatic mitochondria isolated from control and intoxicated mice (Figure 3B).
