2022, International Journal of PharmaceuticsCitation Excerpt :Regarding the efficient delivery of antiretrovirals from the circulating blood to the testis, it is of particular importance to understand the characteristics of the AZT transport mechanism at the BTB. Carrier-mediated transport by influx transporters expressed at the BTB are considered to be involved in the AZT transport mechanism, although the distribution of AZT was reported to be in a passive diffusion manner (Hu, 1993, Masereeuw et al., 1994). AZT is a nucleoside analog with azide (N3–), and several investigations have been taken to examine the recognition of AZT by transporters, such as the equilibrative nucleoside transporters (ENT2,3/solute carrier (SLC) 29A2,3) (Yao et al., 2001, Rahman et al., 2018), the concentrative nucleoside transporters (CNT1,3/SLC28A1,3) (Yao et al., 2001), the organic anion transporters (OAT1,2,3,4/SLC22A6,7,8,11) (Wada et al., 2000, Morita et al., 2001, Takeda et al., 2002), and the organic cation transporter 1 (OCT1/SLC22A1) (Chen and Nelson, 2000).

This result could be correlated to the high lipophilicity of the prodrug (Table 3); AZT has been described to cross Caco‐2 monolayers mainly by a passive diffusion process.41 Despite relative stability in Caco‐2 cell homogenate compared with CEM‐SS cell extract (Table 2), extensive metabolism of the tBuSATE prodrug occurred, the intact prodrug 2 constituting < 10% of the AZT equivalents measured in the acceptor compartment.
