Cells were incubated with 10 nM insulin for 60 min at 37°C. After drug treatment, uptake and binding assays in intact SK-N-SH cells were carried out as described under Materials and Methods. Km and Vmax values of transport kinetics were derived from Hofstee modification of the Michaelis-Menten equation whereas Kd and Bmaxvalues of binding assays were derived from Scatchard analysis. Data are presented as mean ± S.E.M. of three experiments carried out in triplicate.
