Rate of PG and 6-MP accumulation in buffer, liver, and kidney homogenate

Rate of Accumulation
PG6-MP
nmol/ml/min pmol/ml/min
Buffer1.12  ± 0.025.4  ± 0.8

Kidney homogenate0.30  ± 0.051-c 38.0  ± 3.41-b

PTA (1 mM), GSH (5 mM), and allopurinol (5 mM) were incubated in the presence of buffer, liver, or kidney homogenate. Protein concentration in liver and kidney homogenate incubations was 2.32 ± 0.03 and 2.50 ± 0.13 mg, respectively. Samples were taken every 15 min and analyzed by HPLC as described in Materials and Methods.Values presented are the mean ± S.D. from three independent experiments.

  • 1-a Significantly different from buffer-only (p < .05).

  • 1-b Significantly different from buffer-only and liver homogenate (p < .05).

  • 1-c No increase in PG accumulation was observed after 15 min in kidney homogenate incubations.
