Membranes were incubated with 50 nM CACO, 50 nM N-CPM-CACO, 8 nM CAM, or 64 nM MC-CAM for 15 min at 25°C. Control samples lacked compound in preincubation. Membranes were then diluted with 50 mM Tris-HCl, pH 7.5, followed by four centrifugal washes. Membranes were finally resuspended in 50 mM Tris-HCl, pH 7.5, and [3H]DAMGO binding at concentrations ranging from 0.025 to 3.2 nM was measured as described in Materials and Methods. Data are expressed as mean value ± S.E.M. for three determinations performed in triplicate.
