The herbal extract of G. japonicum was fractionated by EtOAc extraction, Sephadex LH-20 column chromatography and HPLC. The fractions separated in each step were examined for anti-HSV activity in the plaque-reduction assay. The cytotoxicity of fraction P4 (eugeniin) was determined by the growth inhibition or [3H]thymidine uptake assay as described in the text. EC50 and CC50values were determined graphically. An asterisk (*) indicates a mean value ± S.D. of three independent experiments. Two asterisks (**) indicate the CC50 value determined by the growth inhibition assay with Vero cells. Three asterisks (***) indicate the CC50value determined by the [3H]thymidine uptake assay with Vero cells. Four asterisks (****) indicate the CC50 value determined by the growth inhibition of HEL cells.
