The reaction mixture contained microsomes (200 μg), 300 μM acetaldehyde and 10 mM NADPH in a final volume of 100 μl, which was incubated for 15 min at 37°C in 0.1 M Tris-acetate buffer, pH 7.2. Acetate was separated from acetaldehyde by anion-exchange column chromatography.
