PACE Y1 - 1973/08/01 UR - http://jpet.aspetjournals.org/content/186/2/315.abstract N2 - Diphenylbarbituric acid was prepared and administered by gavage to male albino SwissWebster mice for evaluation of anticonvulsant, neurotoxic and lethal effects. In these studies the test compound was compared with phenobarbital and diphenylnydantoin, two standard anticonvulsant agents. Diphenylbarbituric acid was effective in blocking the tonic hindlimb extensor phase of maximum electroshock seizures and also protected mice against the clonic seizures and death produced by pentylenetetrazol. Because of limited aqueous solubility, gastrointestinal absorption was not efficient and neurotoxicity, as determined by a rotorod technique or lethality, could not be achieved. When administered intraperitoneally, diphenylbarbituric acid is effective by two standard laboratory tests used for the demonstration of anticonvulsant efficacy; moreover, by this route, estimates of neurotoxic and lethal doses were made. © 1973 by The Williams & Wilkins Co.
