PLETSCHER Y1 - 1971/07/01 UR - http://jpet.aspetjournals.org/content/178/1/210.abstract N2 - In blood platelets of man, rabbits, guinea pigs and cattle, in isolated storage organelles of rabbit and guinea-pig platelets as well as in chromafl in granules of bovine adrenal medulla the content of 5'-phosphonucleotides has been determined. The main part of the nucleotides of both platelets and storage organelles (80-87%) consisted of derivatives of adenosine [adenosine-5'-triphosphate (ATP), adenosine-5'-diphosphate (ADP) and adenosine-5'-monophosphate (AMP)] whereas the rest derived from guanosine [guanosine-5'-triphosphate (GTP), guanosine-5'-diphosphate and guanosine-5'-monophosphate (GMP)] and to a small extent from uridine (uridine-5'-triphosphate). The concentration in total 5'-phosphonucleotides (in nanomoles per microgram of protein) of isolated platelet organlles was 200 to 300 times higher than that of the corresponding platelets. Storage organelles contained almost exclusively tri- and diphosphonucleotides whereas in the whole platelets an appreciable proportion of monophosphonucleotides was present in addition. The ATP/ADP quotient of platelets and/or organelles was low in man and guinea pigs but high in rabbits. Bovine platelets showed an intermediate value. Chromaffin granules behaved like the storage organelles of rabbits but had a lower concentration of total 5'-phosphonucleotides than the latter. From these and previous findings it is concluded that in subcellular organelles ATP, ADP and GTP in contrast to AMP and GMP are probably involved in the storage of monoamines. © 1971, by The Williams & Wilkins Company ER -
