It has been shown that after administration of a single dose of mephobarbital to rats the anticonvulsant response seen up to three hours can be ascribed to mephobarbital. At four hours and later a significant concentration of phenobarbital, the demethylated metabolic product of mephobarbital, was present in the brain and presumably contributed to the anticonvulsant activity. The extent to which phenobarbital contributes to the anticonvulsant activity of mephobarbital with long-term administration of the latter cannot be stated on the basis of these studies. © 1971, by The Williams & Wilkins Company ER -
