Mattis Y1 - 1966/06/01 UR - http://jpet.aspetjournals.org/content/152/3/509.abstract N2 - Time effects of doses of probenecid from 25 to 100 mg/kg on the hepatic uptake, storage and removal of indocyanine green (ICG) were studied in unanesthetized and anesthetized beagles. In the unanesthetized dog, pretreatment with probenecid was found to decrease tile plasma clearance rate of dye in a dose-related manner. Administration of probenecid following ICG injection resulted in a hyperbolic plasma clearance curve. From time asymptotes of this curve, it was possible to estimate the rates of transfer between plasma, liver and bile. In animals receiving a continuous infusion of ICG, prohenecid was shown to increase plasma concentration of dye, decrease biliary ICG content and increase bile flow.
