YR 1962 UL http://jpet.aspetjournals.org/content/138/2/258.abstract AB An ultraviolet spectrophotometric method was developed in this laboratory to follow the metabolism of 1-methyl-2-formylpyridinium iodide oxime (2-PAM) in liver perfusates. This determination was based on the bathochromic shift of 2-PAM in alkaline solutions. Approximately 0.01 µmole per ml of 2-PAM can be determined by this method. Formation of the zwitterionic species of 2-PAM was established as the form of 2-PAM which produced the bathochromic displacement. Carbon-14 labeled 2-PAM was prepared and its biotransformation was also investigated in the isolated perfused rat liver. Both ultraviolet absorption analyses and isotopic tracer studies indicated that extensive biotransformation of 2-PAM had occurred.
