These results were compared to a single intravenous injection of guanethidine to intact dogs with cannulation of the adrenal vein and measurement of norepinephrine and epinephrine secretion rates. Direct perfusion of the isolated denervated adrenal gland preparation with guanethidine and histamine resulted in no effect on catecholamine secretion. Blood vessel tonus, as manifested by constant mean perfusion pressure, was unaffected by guanethidine indicating no direct action of the drug on vessel walls in this type of preparation. Guanethidine in a single intravenous dose to the intact dog preparation likewise resulted in no adrenal medullary stimulation. However, the epinephrine secretion rate was decreased by greater than 50%. ER -
