The analgesic efficacy and side action liability of nalorphine and nalorphinemorphine combinations have been studied in postoperative patients and healthy volunteers. 2. Nalorphine, in 5 mgm. doses, produces side effects comparable to those produced by 10 mgm. of morphine, but is devoid of significant analgesic activity. In higher doses, nalorphine has powerful analgesic action, but frequently produces unpleasant side effects. Since most. workers have failed to find significant analgesic effect with nalorphine in animals, our data suggest a significant species difference in this regard. 3. In our studies the combination of 2 mgm. of nalorphine and 10 mgm. of morphine produced analgesia and side effects indistinguishable from those achieved with 10 mgm. of morphine. 4. The combination of 5 mgm. of nalorphine and 15 mgm. of morphine produced respiratory depressant and subjective side effects similar to those obtained with 15 mgm. of morphine. 5. The difficulties involved in the formulation of clinically useful nalorphine-morphine mixtures are discussed. ER -
