Fig. 2. 1-13C glucose label flow diagram. Relative carbohydrate versus FFA/ketone oxidation assessed as the relative contribution
of each of these substrate sources to acetyl-CoA metabolism can be determined in myocardial tissue extracts following the
in vivo steady state administration of 1-13C-glucose. The diluted 13C label entering into the 2-13C acetyl CoA pool through
PDH is a result of carbohydrate oxidation including glucose, glycogen, lactate, and pyruvate. The degree to which the 4-13C
glutamate pool is diluted from the 3-13C pyruvate (equivalent to the 3-13C lactate and 3-13C alanine pool enrichment at steady
state) pool, depends on the extent of label dilution from unlabeled FFA and ketone body oxidation present. Therefore, the
relative carbohydrate oxidation (%) is defined as: {4-13C-glutamate/(3-13C- lactate + 3-13C- alanine)/2}100 and relative
FFA/ketone oxidation (%) is:100-relative carbohydrate oxidation.
Cit, Citrate; Mal, Malate; Pyr, pyruvate; Asp, aspartate; PDH, pyruvate dehydrogenase.
