Effect of modulators of the Erk1/2 signaling pathway on AA-mediated phosphorylation of Erk1/2 (A) and Raf (B). Forty-eight hours after plating, hepatocytes were treated with AA alone (5 mM) or with various modulators of Erk1/2 signaling, Genistein (Gen, 50 μM), B581 (50 μM), PD98059 (PD, 50 μM), and bisindolylmaleimide (Bis, 5 μM), 1.5 h before the addition of AA. p-pErk1/2 or p-Raf levels were monitored by immunoblot analysis and band density normalized to total Erk1/2 or Raf levels, respectively. pErk1/2 or p-Raf levels are plotted as a percentage of the p-Erk1/2 or p-p-Raf level monitored in untreated hepatocytes.
