Abstract
Desmethylimipramine effectively inhibited basal (interdigestive) gastric acid secretion in the rat at p.o. doses of 4 to 50 mg/kg. The drug also inhibited acid secretion induced by 2-deoxy-D-glucose in the conscious gastric fistula dog at i.v. doses of 2.5 and 5.0 mg/kg. These doses, however, were ineffective in inhibiting carbachol-induced acid secretion in the Heidenhain pouch dog. Additionally, the 5-mg/kg dose did not block the acid secretion induced by electrical preganglionic vagal stimulation in the anesthetized dog. The results suggest that the gastric antisecretory action of desmethylimipramine is mediated at a site in the central nervous system.
