Abstract
During a constant rate infusion of C14-tryptophan, the specific activity of plasma tryptophan and brain 5- hydroxytryptamine increased curvilinearly with time. Since the steady-state concentration of plasma tryptophan (TP) and brain 5-hydroxytryptamine (5-HT) remained unchanged during the infusion, the fractional rate constant for TP (kTP) and brain 5-HT (k5-HT) could be calculated by applying concepts of steady-state kinetics. The product of k5-HT and the brain 5-HT concentration is a measure of the turnover rate of brain 5-HT. The values calculated with this method approximate the turnover rate values obtained with nonisotopic methods.
