Abstract
The influence of hemicholinium (HC-3) on the in vitro uptake of C14-choline by the rabbit heart was studied. The hearts were perfused 1 hr in an oxygenated recycling system at 37°c with eserine-treated Locke-Ringer's solution, containing 0.012 µml of C14-choline and 50 µg/ml of HC-3. Control hearts received no HC-3. Maximum uptake occurred in 60 min. Immediately after perfusion, the hearts were ground in liquid nitrogen and the C14-labeled compounds were quantitatively extracted (control recoveries of 94 ± 3%) and separated by paper chromatography. The results indicated a marked reduction in C14-choline uptake in the presence of HC-3 (62 ng/g vs. 24 ng/g). No incorporation of the C14-label into acetylcholine was observed.
