1. The compound, 3-acetoxy phenyltrimethylammonium methylsulfate (Nu 2017), produces contractile responses in acutely and chronically denervated mammalian muscle like those produced by acetylcholine.
2. Nu 2017 enhances the isometric response of the muscle to single maximal nerve stimuli. The augmented mechanical response is associated with repetitive electrical activity in the muscle.
3. The action of Nu 2017 on the neuromuscular mechanism is intensified by the prior inactivation of cholinesterase.
4. After initial stimulation large doses of Nu 2017 produce a transient depression of neuromuscular function.
5. The anti-curare activity of Nu 2017 is a prominent feature of its action on neuromuscular function.
6. The activity of several cholinergic compounds on neuromuscular function is discussed in relation to the effects of Nu 2017 that are qualitatively similar to those of neostigmine and acetylcholine. These effects of Nu 2017 may be summarized as equivalent to an abbreviated neostigmine action and a moderately prolonged acetylcholine action.
