1. A method for the assay of the oxytocic principle in pituitary extract (posterior lobe) is described.
2. Eight strips of muscle from the uterus of one guinea-pig suspended in Van Dyke and Hastings' solution are used for the test.
3. Potencies are calculated in terms of a standard from regression lines computed on the basis of an all-or-none response.
4. Selection of guinea-pigs for the test is simplified since weight within wide limits and the stage in the oestrous cycle are not important. All guinea-pigs used gave satisfactory assays.
5. The average error of 26 individual assays was 5.98 ± 7.13 per cent.
6. The errors of each individual test may be computed independently.
7. The results from a number of samples assayed by this method are reported.
