ALKYL NITRITES VI. A CONTRIBUTION TO THE MECHANISM OF THE ACTION OF ORGANIC NITRATES

JOHN C. KRANTZ JR. 1, C. JELLEFF CARR 1, SYLVAN E. FORMAN 1, and NELLIE CONE 1

1 From the Department of Pharmacology, School of Medicine, University of Maryland, Baltimore, Maryland

1. Evidence has been produced to indicate that the action of certain organic nitrates is not dependent upon their hydrolysis and nitrite formation, but rather upon the unhydrolyzed molecule.

2. The presence of a carboxyl group in the molecule of certain organic nitrates does not diminish the depressor response if a high oil/water coefficient is maintained.

3. The tolerance upon intravenous injection acquired by the dog to water-soluble organic nitrates appears to be far greater than that produced by the water-insoluble compounds.
